Protective effects of piperine on the retina of mice with streptozotocin-induced diabetes by suppressing HIF-1/VEGFA pathway and promoting PEDF expression被引量:4

AIM:To evaluate the protective mechanisms of piperine in the retina of mice with streptozotocin-induced diabetes.METHODS:In experiments in vitro,stimulation by chemical hypoxia was established in ARPE-19 cells.Then,the expression of hypoxia-inducible factor-1α(HIF-1α),vascular endothelial growth factor A(VEGFA),and pigment epithelium-derived factor(PEDF)was assessed at the m RNA and protein levels.In experiments in vivo,diabetes mellitus was established by intraperitoneally injecting 150 mg/kg streptozotocin once.After 3 wk of the onset of diabetes,15 mg/kg piperine was intraperitoneally injected once daily for 1 or 3 wk.Then,the retinal morphology and m RNA and protein expression were assessed.RESULTS:In hypoxia,1-100μmol/L piperine significantly decreased the expression of VEGFA m RNA and increased the expression of PEDF m RNA without affecting HIF-1αm RNA.Meanwhile,100μmol/L piperine substantially decreased the protein level of VEGFA and increased the protein level of PEDF.The HIF-1αprotein level was also hampered by piperine.In the diabetic retina of mice,the morphological damage was alleviated by piperine.Likewise,the retinal vascular leakage was substantially decreased by piperine.Further,the protein levels of HIF-1αand VEGFA were significantly reduced by piperine.Moreover,the level of the antiangiogenic factor of PEDF dramatically increased by piperine.CONCLUSION:Piperine may exert protective effects on the retina of mice with diabetes via regulating the pro-antiangiogenic homeostasis composed of HIF-1/VEGFA and PEDF.

糖尿病患者视网膜神经节细胞复合体和神经纤维层厚度的变化被引量:4

目的观察2型糖尿病(T2DM)患者中视网膜神经纤维层(RNFL)和神经节细胞复合体(GCC)厚度的变化,分析其神经结构与微循环之间的相关关系。方法采用横断面回顾性研究,入选2型糖尿病患者45例71眼、健康受试者36例68眼。采用光学相干断层扫描血管成像技术(OCTA)检测黄斑部的GCC厚度、视盘周围RNFL厚度、视网膜脉络膜毛细血管丛的微血管密度。观察不同分期糖尿病视网膜病变(DR)患眼中RNFL和GCC厚度变化,分析它们与微血管密度的相关关系。结果与对照组相比,NDR组(无DR的糖尿病患者)GCC中局部丢失体积(FLV,P=0.00)、整体丢失体积(GLV,P=0.00)显著升高,可能是视网膜神经结构损伤的早期敏感指标。与对照组相比,轻中度非增殖性糖尿病视网膜病变(NPDR)组的下侧和鼻侧象限RNFL显著变薄(P=0.00),可能是RNFL丢失的敏感区域。重度NPDR组中鼻侧象限RNFL仍然显著变薄,但下侧、颞侧象限RNFL显著增厚(P=0.00)。增殖性糖尿病视网膜病变(PDR)组中RNFL和GCC大部分参数普遍增厚(P=0.00),可能与视网膜水肿导致的视网膜增厚有关。Spearman分析显示GCC、RNFL厚度与视网膜厚度呈正相关。此外,GCC及RNFL厚度与视网膜脉络膜毛细血管包括浅层毛细血管丛(SCP)、深层毛细血管丛(DCP)和脉络膜层毛细血管丛(CCP)的密度呈负相关。结论GCC的FLV和GLV局限性丢失,可能是评价早期视网膜神经结构损伤的敏感指标。随DR的进展,RNFL厚度呈现先变薄后增厚的趋势,还需要扩大样本量进一步研究。

视盘玻璃疣的研究现状及进展被引量:4

视盘玻璃疣(ODD)是无细胞性沉积物,位于视盘的筛板前部。大多数患者无明显临床症状,通常被忽视,也容易与全身高危疾病导致的视盘水肿相混淆。目前主流观点认为,视神经纤维轴突代谢紊乱,导致细胞内线粒体钙化,轴突慢性崩解后钙化的线粒体不断释放到细胞外,导致细胞外钙浓度远较细胞内高,钙质不断积聚从而产生微小的钙化体,多个钙化体逐渐融合形成ODD。增强深度成像OCT能敏感地检测ODD,其影像特征是被强反射边缘完整或部分包绕的弱反射核心。ODD是造成视盘拥挤的重要原因,在青春期,埋藏型玻璃疣逐渐发生钙化,向视盘浅表迁移,转变为浅表型ODD,因而部分ODD患者在青春期突然进展,成年期趋于稳定,可以伴发视野缺损、前部缺血性视神经病变等血管并发症。

MiR-200c suppresses the migration of retinoblastoma cells by reversing epithelial mesenchymal transition被引量:3

AIM: To analyze the relationship between clinical features and epithelial mesenchymal transition (EMT) in retinoblastoma (RB), further to investigate whether miR-200c regulates the EMT and migration of RB cells. METHODS: Expression of EMT-related markers and tumor- related factors were detected by immuno-histochemistry analysis in RB tissue from 29 cases. Correlations between their expression and clinical characteristics were analyzed. The regulation effects of miR-200c on EMT-related markers, tumor-related factors were observed in mRNA level and protein level by real-time polymerase chain reaction (PCR) and Western blot, respectively, in Y79 and Weri-rbl cells. Its effects on migration force of these RB cell lines were also detected with Transwell test. RESULTS: Lower expression of E-cadherin was present in the cases with malignant prognosis. MiR-200c promoted the expression of E-cadherin and decreased the expression of Vimentin and N-cadherin in Y79 and Weri-rbl cells. Migration force of RB cells could be inhibited by miR-200c. CONCLUSION: EMT might be associated with bad prognosis in RB. MiR-200c suppresses the migration of retinoblastomatous cells by reverse EMT.
