3'-端肽缀合修饰提高寡核苷酸的血清稳定性(英文)

RNA干扰被广泛用于传染性疾病及恶性肿瘤的基因治疗领域,但体内稳定性较差限制了其在临床上的应用。本文报道了在siRNA正义链的3'-端缀合不同长度的肽片段均能够明显提高血清稳定性,而在siRNA反义链的3'-端缀合相同的肽片段则对血清稳定性没有影响。血清中的RNaseA能够选择性水解siRNA正义链3'-末端缀合的肽片段,从而得到原siRNA片段,但反义链3'-末端缀合肽缀合物则不能在血清中得到原siRNA片段。该结果进一步证明RNase A对siRNA双链的进攻具有方向性。

Ficolins are serum complement lectins,with a structure similar to mannose-binding lectin(MBL) and lung surfactant protein(SP)-A and SP-D.Ficolins activate the lectin complement system and play important roles in host innate immunity.Ficolins are members of the collectin family of proteins,which act as pattern recognition receptors(PRRs).They are soluble oligomeric defense proteins with lectin-like activity,and are able to recognize pathogen-associated molecular patterns(PAMPs),which are carbohydrate molecules on the surface of pathogens,and of apoptotic,necrotic,and malignant cells.Upon binding to their specific PAMPs,ficolins may trigger activation of the immune system either(1) by initiating activation of complement via the lectin pathway,(2) by a primitive type of opsonophagocytosis,or(3) by stimulating secretion of the inflammatory cytokines interferon(IFN)-y,interleukin(IL)-17,IL-6,and tumor necrosis factor(TNF)-a,and production of nitric oxide(NO)by macrophages,thus limiting the infection and concurrently orchestrating the subsequent adaptive immune response.Recently,a number of reports have shown that dysfunction or abnormal expression of ficolins may play crucial roles in viral and bacterial diseases and in inflammation.This review summarizes the reports on the roles of ficolins in the infectious diseases,and provides insight into ficolins as novel innate immune therapeutic options to treat these diseases.

Tumor Carbohydrate Antigens and Strategies to Develop Cancer Vaccines and Drugs

Certain carbohydrate antigens of malignantly trans- formed cells have been identified as markers for the onset of can- cer and have become targets for the development of anticancer vaccine therapies. For tumor antigens, many carbohydrate antigens belong to T-independent (TI) antigens. Carbohydrate conjugated to protein carriers can switch TI antigen to a T-dependent (TD) anti- gen. Attempts to add an innate immune response element (such as Toll-like receptor ligand) to carbohydrate TI-antigens have also been studied. Glycosylation inhibitors or small interfering RNA have also been used for antitumor and/or antiviral agents. This review aims at describing the vast spectrum of tumor carbohydrate antigens and strategies to develop cancer vaccines and drugs.

小干扰RNA(small interfering RNA,si RNA)能够对靶m RNA进行高效性、高特异性转录后调控,具有可传递性等特点,已被用于多种疾病的治疗研究.但其自身存在的诸多缺陷(如稳定性差、难以跨膜等)限制了它在临床上的广泛应用.对核苷酸单元进行结构改造,或构建缀合物,均可以在一定程度上提高si RNA的成药性.本文针对各类si RNA缀合物的研究进展进行了系统总结,阐述了各自独特的生物学特点及潜在的不足,并对其临床应用前景进行了展望.

丙型肝炎病毒感染人肝细胞系对多种唾液酸转移酶和半乳糖基转移酶表达的影响被引量:1

目的探讨丙型肝炎病毒(hepatitis C virus,HCV)感染人肝癌细胞系Huh7.5.1细胞后与肿瘤相关的唾液酸转移酶家族和半乳糖基转移酶家族的35个糖基转移酶的表达变化。方法利用HCV感染的Huh7.5.1肝细胞模型,采用Western blot检测HCV感染后HCV非结构蛋白NS3的表达,定量即时聚合酶链锁反应(quantitative real-time polymerase chain reaction,qRT-PCR)检测HCV感染后HCV-RNA的表达及HCV感染对Huh7.5.1细胞中35个糖基转移酶mRNA表达水平的影响。凝集素染色实验测定HCV感染对Huh7.5.1细胞中糖苷表达水平的影响。结果在HCV感染后的Huh7.5.1细胞中,发现4个唾液酸转移酶(ST3Gal III、ST6Gal NAC III、ST8Sia III和ST8Sia IV)和5个半乳糖基转移酶(B3GALT 1、B3GALT 2、B3GALT 3、B3GALT 4和B4GALNT 2)的mRNA水平明显升高11~45倍,其中变化最大的是B3GALT 1,上升45倍,其次是ST8Sia III和ST8Sia IV,分别上升37倍和39倍。进一步采用凝集素染色实验验证,发现结合末端唾液酸糖苷Neu5Acα6Gal的接骨木凝集素(elderberry lectin,EBL)在HCV感染后升高1.97倍,结合末端半乳糖苷Galβ1,3Gal NAc的尾穗苋凝集素(amaranthus caudatus agglutinin,ACA)和结合Galβ1,4Gal NAc的鸡冠刺桐凝集素(erythrina cristagalli agglutinin,ECA)分别升高3倍和4.3倍。结论 HCV感染导致人肝细胞中特定唾液酸转移酶和半乳糖基转移酶及其相应糖苷表达水平呈现明显上升。这些结果对研究HCV感染的治疗靶点和诊断标志物具有一定参考价值。

Synthesis and biological evaluation of peptide-siRNA conjugates with phosphodiester unit as linker

In this paper,a series of peptide-siRNA conjugates with phosphodiester unit as the linker targeting to Cdc2 gene were synthesized by solid phase stepwise strategy.The conjugation of peptides at either 3’-terminus of siCdc2 bring no change to the classical A-form of RNA duplex,but slightly compromise the thermodynamic stability.Peptide conjugation at the 3’-terminus of sense strand could improve the serum stability obviously,however,the opposite peptide conjugation at the 3’-terminus of antisense strand shows no such influence.According to the results of artificial silencing activity assay system,peptide conjugation at 3’-terminus of antisense strand slightly weakens the silencing activity of siCdc2.But sense strand peptide conjugation exhibits similar silencing activity as native siCdc2,meanwhile,it could mitigate the unwanted off-target effect of sense strand targeting to its own mRNA.

Loss of silencing activity caused by 5′-terminal modification with D-/L-isonucleotide(isoNA) or locked nucleic acid(LNA) could not be restored by 5′-terminal phosphorylation被引量:1

In this study,a series of 5′-phosphorylated siRNAs with D-/L-isonucleotide(isoNA)or locked nucleic acid(LNA)incorporated at the 5′-terminus were synthesized.It was found that after incorporating either isoNA or LNA at the 5′-terminus of the antisense strand or sense strand,the silencing activity of modified strands has been inhibited,which cannot be recovered by phosphorylation at the 5′-terminus;however,the silencing activity of unmodified strand to its own target was increased.This work indicates that the isoNA and LNA modification at 5′-terminus can interfere with the strand selection during the RISC assembling process,and the disturbance of the 5′-phosporylation should not be the only viable mechanism.

利用共聚焦显微镜研究siRNA/CLDs复合物的胞内分布(英文)

研究siRNA在胞内的运输过程有助于阐明其在细胞质基质中的动力学性质及过程。本文报道了激光共聚焦扫描显微镜技术用于研究siRNA及其肽缀合物与胱氨酸骨架阳离子脂质体递送系统(siRNA/CLD)的胞内分布过程。发现siRNA在胞质内分布动力学与3′-单肽-siRNA缀合物/CLD递送系统的入胞途径紧密相关,其中具有较好抗肿瘤活性的3′-p As-siRNA/CLD,具有较快的细胞摄取及溶媒体逃逸过程,并能在A375细胞中潴留较长的时间。该方法高效、可定量并可视化,不仅能够测定siRNA在胞内的浓度,也能够定量分析其与细胞亚结构的共定位关系,进一步可系统研究siRNA及其缀合物在胞质内的动力学分布过程。

茎环引物实时定量PCR技术用于检测异核苷修饰的siRNA的可行性研究被引量:1

本文考察了茎环引物实时定量PCR技术用于定量检测D-/L-异核苷(D-/L-iso NA)修饰的siRNA的可行性.根据阈值循环数(Ct值)及异核苷修饰的siRNA模板的初始浓度的对数值(lgC)绘制了标准曲线,与siRNA的标准曲线进行对比,发现D-异尿苷(D-isodU)修饰的siRNA(D-isodU-siRNA)不影响茎环引物实时定量PCR过程,而L-isodU修饰的siRNA(L-isodU-siRNA)降低了逆转录效率,而使测得的Ct值偏高,且对逆转录效率的影响有浓度依赖性和修饰位点特异性.因此,对前者既可进行绝对定量也可进行相对定量,对于后者只能进行绝对定量.将上述方法应用于isodU-siRNA血清稳定性的检测,获得了更可靠的定量结果.运用双标准曲线法,可进行胞内isodU-siRNA的定量检测.
