Heterologous gene expression driven by carbonic anhydrase gene promoter in Dunaliella salina

Dunaliella salina, a halotolerant unicellular green alga without a rigid cell wall, can live in salinities ranging from 0.05 to 5 mol/L NaC1. These features of D. salina make it an ideal host for the production of antibodies, oral vaccine, and commercially valuable polypeptides. To produce high level of heterologous proteins from D. salina, highly efficient promoters are required to drive expression of target genes under controlled condition. In the present study, we cloned a 5' franking region of 1.4 kb from the carbonic anhydrase (CAH) gene ofD. salina by genomic walking and PCR. The fragment was ligated to the pMD18-T vector and characterized. Sequence analysis indicated that this region contained conserved motifs, including a TATA- like box and CAAT-box. Tandem (GT)n repeats that had a potential role of transcriptional control, were also found in this region. The transcription start site (TSS) of the CAH gene was determined by 5' RACE and nested PCR method. Transformation assays showed that the 1.4 kb fragment was able to drive expression of the selectable bar (bialaphos resistance) gene when the fusion was transformed into D. salina by biolistics. Northern blotting hybridizations showed that the bar transcript was most abundant in cells grown in 2 mol/L NaCl, and less abundant in 0.5 mol/L NaCl, indicating that expression of the bar gene was induced at high salinity. These results suggest the potential use of the CAH gene promoter to induce the expression of heterologous genes in D. salina under varied salt condition.

To develop an economical and feasible approach to probe protein complexes, differential centrifugation and three-dimensional polyacrylamide gel electrophoresis (PAGE) were performed to separate protein complexes from the cell lysate of human pancreatic cancer cell line, SW1990, followed by mass spectrometric identification. Four macromolecular protein complexes were separated and identified unambiguously.

Intelligent Detection and Identification in Fiber-Optical Perimeter Intrusion Monitoring System Based on the FBG Sensor Network被引量:4

A real-time intelligent fiber-optic perimeter intrusion detection system (PIDS) based on the fiber Bragg grating (FBG) sensor network is presented in this paper. To distinguish the effects of different intrusion events, a novel real-time behavior impact classification method is proposed based on the essential statistical characteristics of signal's profile in the time domain. The features are extracted by the principal component analysis (PCA), which are then used to identify the event with a K-nearest neighbor classifier. Simulation and field tests are both carried out to validate its effectiveness. The average identification rate (IR) for five sample signals in the simulation test is as high as 96.67%, and the recognition rate for eight typical signals in the field test can also be achieved up to 96.52%, which includes both the fence-mounted and the ground-buried sensing signals. Besides, critically high detection rate (DR) and low false alarm rate (FAR) can be simultaneously obtained based on the autocorrelation characteristics analysis and a hierarchical detection and identification flow.

Multi-scale wavelet analysis of TOPEX/Poseidon altimeter significant wave height in eastern China seas

The multi-scale characteristics of wave significant height (Hs) in eastern China seas were revealed by multi-scale wavelet analysis. In order to understand the relation between wave and wind, the TOPEX/Poseidon measurements of Hs and wind speed were analyzed. The result showed that Hs and wind speed change in multi-scale at one-, two-month, half-, one- and two-year cycles. But in a larger time scale, the variations in Hs and wind speed are different. Hs has a five-year cycle similar to the cycle of ENSO variation, while the wind speed has no such cycle. In the time domain, the correlation between Hs and ENSO is unclear.

Changes in Huanghe (Yellow) River estuary since artificial re-routing in 1996被引量:4

With the combination of historical data, field observations and satellite remotely sensed images (Landsat TM/ ETM+ and CBERS), changes in Huanghe (Yellow) River estuary since 1996 when artificial Chahe distributary was built up were studied, mainly including water and sediment discharge from the river, tides, tidal currents, suspended sediment diffusion, coastline changes and seabed development. During following six and half years (up to the end of 2002), runoff and sediment loads into the river mouth declined dramatically. At the beginning of the re-routing, abundant sediment loads from the river filled up nearshore shallow water areas so that the newborn delta prograded quickly. With rapid decrease of sediment loads transported to the estuary, the delta retrograded. In 1997, subaerial tip of the abandoned delta receded 1.5km; its annual mean recession rate was about 150 m in following years. In addition, marine dynamic condition near the artificial outlet had also changed. Under the interaction of ocean and river flow, most of incoming sediment loads deposited in the vicin- ity of the outlet. Seabed erosion occurred at the subaqueous delta front. Between 1999 and 2002, erosion thick- ness averaged at 0.3 m in the subaqueous delta of 585.5 km2.

MECHANICAL-ELECTRIC COUPLING DYNAMICAL CHARACTERISTICS OF AN ULTRA-HIGH SPEED GRINDING MOTORIZED SPINDLE SYSTEM被引量:23

On the basis of the traditional mechanical model of a grinding wheel rotor and the mechanical-electric coupling model with ideal sinusoidal supply, taking high-frequency converting current of inverter power switches into further consideration, a modified mechanical-electric coupling model is created. The created model consists of an inverter, a motorized spindle, a grinding wheel and grinding loads. Some typical non-stationary processes of the grinding system with two different supplies, including the starting, the speed rising and the break in grinding loads, are compared by making use of the created model. One supply is an ideal sinusoidal voltage source, the other is an inverter. The theoretical analysis of the high-order harmonic is also compared with the experimental result. The material strategy of suppressing high-order harmonic mechanical-electric coupling vibration by optimizing inverter operating parameters is proposed.

三阶多粒度光交叉连接的优化设计研究(英文)

在采用密集波分复用技术的骨干网中,通过建立动态多粒度业务模型对由大粒度(光纤和波带)和小粒度(波长)组成的三阶多粒度光交叉连接所构成的节点的网络特性(阻塞概率和带宽阻塞比)进行了研究和仿真分析,并由此对三阶多粒度节点的结构参数和网络中的系统参数进行了优化设计.从仿真结果可以看出,所提出来的优化设计方案能够显著的减少节点结构的复杂度并改善阻塞性能,从而减小了网络中设备的规模和成本.
