硒化合物诱导人高转移大细胞肺癌L9981产生活性氧变化的研究被引量:2

背景与目的已有研究表明甲基硒酸具有防癌和抗癌作用。本研究拟探讨甲基硒酸及亚硒酸钠在人高转移大细胞肺癌L9981的代谢过程中是否产生活性氧及其变化,进一步明确氧化应激在不同硒化合物中发挥抗癌效应的地位。方法应用细胞活力计数仪,检测硒化合物对L9981的生长抑制影响;采用活性氧荧光探针羧基-H2DCFDA探测经硒化合物诱导的L9981细胞内活性氧的含量;采用N-乙酰半胱氨酸(NAC)与硒化合物同时处理L9981细胞后,流式细胞术检测细胞内活性氧的变化。结果①2.5μMMSA即可明显抑制L9981细胞生长,且随浓度增加,效应增强。②甲基硒酸及亚硒酸钠均可诱导L9981产生显著的ROS变化,亚硒酸钠产生时间早于MSA;③采用NAC同时处理L9981后,甲基硒酸能够显著的促进细胞产生活性氧,而亚硒酸钠能够显著的抑制细胞内活性氧的产生。结论①甲基硒酸及亚硒酸钠对L9981均具有抑制生长的作用,并且具有剂量依赖性;②氧化应激反应参与了MSA及亚硒酸钠诱导L9981细胞死亡的过程。
