颅内压增高皮层脑电的复杂度分析被引量:3

为研究颅内压增高对脑功能的影响,以家兔为实验对象建立侧脑室灌注加压的单纯颅高压模型.采集皮层脑电信号并同步记录颅内压(ICP)、心电、呼吸、动脉血压等信号,采用迟滞粗粒化方法和Lempel-Ziv算法计算脑电信号的复杂度,分析不同ICP状态下脑电信号的非线性变化特征.发现以灌注加压前基础ICP时的状态为对照,ICP为2,4倍基础值时的脑电信号复杂度显著降低(P<0.01);ICP越高,复杂度越低;ICP恢复正常后,脑电复杂度有恢复趋势.研究结果表明,脑电信号的复杂度可用于检测由于ICP增高引起的脑损伤并有可能对其损伤程度进行定量评估.

神经元兴奋性群体峰电位信号的Renyi信息表达被引量:5
2004年
应用时频分布级数分析方法,提取了单次诱发群体峰电位(PS)信号的Renyi信息,并用以表征神经元的兴奋性变化.采用液压打击大鼠脑损伤模型和细胞外记录技术,记录了离体海马脑片CA1区锥体神经元PS,发现损伤侧和非损伤侧PS的Renyi信息参数值差异显著,进而分析了神经元灌流大黄酸后损伤侧PS的Renyi信息参数值的变化,研究大黄酸对神经元的超兴奋性和突触传递的作用.研究结果表明,颅脑损伤可造成海马CA1区锥体神经元的迟发性过度兴奋,大黄酸对神经元的过度兴奋有抑制作用,Renyi信息可作为反映神经元兴奋性变化的一个特征参数.

中枢兴奋性传递的突触后电位小波熵分析研究被引量:7

采用细胞内记录方法提取了离体大鼠海马脑片CA1区锥体神经元兴奋性突触后电位(EPSP);应用小波熵研究了EPSP所包含的信息.在计算了EPSP的小波熵、幅度、上升和下降相速率等参数并对其进行相关矩阵分析后,发现EPSP小波熵与其幅度、上升和下降相速率等参数均具有较强的正相关关系,是EPSP最具代表性的一个特征参数.进而分析了大黄酸对中枢兴奋性传递的作用,发现神经元在灌流大黄酸前后小波熵参数值变化显著.研究表明,小波熵能较全面地表征EPSP信号特征,比传统参数更具代表性,并能较灵敏地反映神经元兴奋性改变,较好地反映神经元间的信息传递.

液压打击损伤后海马CA1区神经元兴奋性变化的研究被引量:7

为考察脑损伤对海马CA1区锥体神经元电活动的影响并研究大黄素对神经元的超兴奋性和突触传递的作用,应用液压打击大鼠脑损伤模型和细胞外记录方法提取诱发的海马CA1区场兴奋性突触后电位(fEPSP)和群峰电位(PS), 进行相关的数据处理和分析。发现损伤侧比非损伤侧的fEPSP斜率明显升高,PS波峰个数显著增加,而PS潜伏期明显减小;在灌流液中施加大黄素,CA1区诱发场电位明显减弱。研究结果表明: 颅脑损伤可造成海马CA1区锥体神经元的迟发性过度兴奋;大黄素对神经元的兴奋性有抑制作用,可能对颅脑损伤后的中枢神经系统具有保护功能。

Biodegradable chitosan scaffolds containing microspheres as carriers for controlled transforming growth factor-β_1 delivery for cartilage tissue engineering被引量:25

Background Natural articular cartilage has a limited capacity for spontaneous regeneration.Controlled release oftransforming growth factor-β_1(TGF-β_1)to cartilage defects can enhance chondrogenesis.In this study,we assessed thefeasibility of using biodegradable chitosan microspheres as carriers for controlled TGF-β_1 delivery and the effect ofreleased TGF-β_1 on the chondrogenic potential of chondrocytes.Methods Chitosan scaffolds and chitosan microspheres loaded with TGF-β_1 were prepared by the freeze-drying andthe emulsion-crosslinking method respectively.In vitro drug release kinetics,as measured by enzyme-linkedimmunosorbent assay,was monitored for 7 days.Lysozyme degradation was performed for 4 weeks to detect in vitrodegradability of the scaffolds and the microspheres.Rabbit chondrocytes were seeded on the scaffolds containingTGF-β_1 microspheres and incubated in vitro for 3 weeks.Histological examination and type Ⅱcollagenimmunohistochemical staining was performed to evaluate the effects of released TGF-β_1 on cell adhesivity,proliferationand synthesis of the extracellular matrix.Results TGF-β_1 was encapsulated into chitosan microspheres and the encapsulation efficiency of TGF-β_1 was high(90.1%).During 4 weeks of incubation in lysozyme solution for in vitro degradation,the mass of both the scaffolds andthe microspheres decreased continuously and significant morphological changes was noticed.From the releaseexperiments,it was found that TGF-β_1 could be released from the microspheres in a multiphasic fashion including aninitial burst phase,a slow linear release phase and a plateau phase.The release amount of TGF-β_1 was 37.4%,50.7%,61.3%,and 63.5% for 1,3,5,and 7 days respectively.At 21 days after cultivation,type Ⅱ collagen immunohistochemicalstaining was performed.The mean percentage of positive cells for collagen type II in control group(32.7%±10.4%)wassignificantly lower than that in the controlled TGF-β_1 release group(92.4%±4.8%,P<0.05).Both the proliferation rateand production o

Effects of rhubarb extracts on hyperexcitability of hippocampal CA1 neurons after fluid percussion injury被引量:2

Objective To investigate the effects of rhubarb extracts,i.e.rhein and emodin,on the neuronal hyperexcitability and synaptic transmission,and to further reveal the mechanism of the secondary brain damage.Methods The fluid percussion injury(FPI) rat model and extracellular recording method were used.The evoked field potentials by stimulating Schaffer collaterals were collected from the ipsilateral(impact side) and the contralateral hippocampal CA1 areas of rat in vitro.And the field potentials,including the field excitatory postsynaptic potential and the population spike,were analyzed.Results After the impact was performed on the rat parietal cortex,the evoked field potentials in the ipsilateral hippocampus CA1 area were enhanced obviously.Rhubarb extracts reduced the slope of the field excitatory postsynaptic potential and the number of the population spike significantly while rhein and emodin increased the latency of the population spike obviously.Conclusion Rhubarb extracts,i.e. rhein and emodin,can depress the neuronal hyperexcitability,which suggests that rhein and emodin play an important role in protecting the central nervous system from neuronal damage after traumatic brain injury.FPI produces hyperexcitability of hippocampal CA1 neurons,probably by enhancing excitatory synaptic transmission.
