Screening of the Gene for Chlorella Identification and Identification of oil-producing Microalgae被引量:2

[Objective] The aim was to select suitable gene for Chlorella identification and to identify the oil-producing microalgae.[Method] Four candidate gene sequences,the nuclear genomic rDNA of the 18S rRNA gene,internal transcribed spacer(ITS),internal transcribed spacer Ⅱ(ITS Ⅱ)and the chloroplast rbcL gene,were selected for Chlorella molecular identification.Through these four candidate genes,the genetic variability and distinguish ability between intra-species and inter-species was analyzed to choose the right genes for identification of the high oil-content Chlorella.On this basis,application of these gene segments were classified and identified for five fresh-water isolated Chlorella,which oil-content is more than 30%.[Result] ITS gene was a suitable gene because of its high variation and short fragment length,meanwhile its genetic distance intra-species(0.439 6±0.135 9)was larger than inter-species(0.045 7±0.084 3).Its sequence length varied between different species whereas highly conserved in the same species.By the application of ITS sequences,respectively,five high oil-content stains were identified as one C.vulgaris,two strains of C.sorokiniana and two strains of algae Chlorella sp.[Conclusion] This study had provided reference for the establishment of identification gene pool of Chlorella.

利用复合微生物除草剂控制稗草和反枝苋的研究被引量:4

本研究利用盆栽试验,对Alternaria augustiovoidea菌株AAEC05-3和A.amaranthi-3开发复合微生物除草剂的潜力进行了评价。结果表明,在不保湿情况下,107个孢子.mL-1浓度的复合菌水乳剂对杂草的防效可达89.15%。复合菌水乳剂与低剂量的化学除草剂混合可以显著提高微生物除草剂对杂草的防除效果,在105个孢子.mL-1浓度下,制剂对反枝苋和稗草防效达到95.56%。

狭卵链格孢菌毒素细交链孢菌酮酸对稗草细胞膜脂质过氧化作用的影响被引量:1

为明确狭卵链格孢菌Alternaria augustiovoide菌株AAEC05-3产生的毒素细交链孢菌酮酸(tenuazonic acid,TeA)的作用机制,利用离体试验,研究了TeA对稗草叶片细胞膜脂质过氧化的影响。结果显示,低浓度TeA处理对稗草叶片的细胞膜透性和脂质过氧化影响不大;用高浓度TeA处理稗草叶片24 h后,稗草叶片细胞膜透性和脂质过氧化作用显著增强。TeA浓度为100μmol/L时,稗草叶片相对电导率和丙二醛(MDA)含量分别增加28.21%和63.58%;脂氧合酶(Lox)活性增高61.59%,且不饱和脂肪酸含量显著降低,亚油酸(18∶2)和亚麻酸(18∶3)分别降低19.56%和17.20%,脂肪酸氢过氧化物在叶片中大量积累。表明TeA可以诱导稗草叶片细胞膜的脂质过氧化作用。

[目的]确定细交链孢菌酮酸(tenuazonic acid,TeA)对稗草的毒害作用是否与其诱导氧化胁迫及抗氧化酶活性变化有关。[方法]通过离体试验研究了TeA处理稗草叶片丙二醛和过氧化氢含量及抗氧化酶活性的变化。[结果]稗草叶片经500μmol/L TeA处理后,叶片中丙二醛和过氧化氢含量分别较对照增加了247.86%和67.00%,表明TeA处理诱导了稗草叶片中活性氧暴发,并导致丙二醛和过氧化氢的大量积累。TeA引起超氧化物歧化酶(SOD)、谷胱甘肽还原酶(GR)和过氧化氢酶(CAT)活性显著升高,其中在500μmol/LTeA处理下,稗草叶片SOD、GR和CAT活性均较对照增加1倍以上。[结论]TeA诱导活性氧的产生引起稗草叶片氧化胁迫,同时诱导叶片抗氧化酶活性升高。

Research on Oxidative Stress Induced by Tenuazonic Acid from Alternaria augustiovoide and Changes in Antioxidant Enzyme Activities in Leaves of Echinochloa crus-galli被引量:5

[Objective] The aim of the study was to determine whether phytotoxicity of TeA against Echinochloa crus-galli leaves was correlated with oxidative stress caused by generation of reactive oxygen and the changes of antioxidant enzymes activity.[Method] The changes of malondialdehyde(MDA)content,hydrogen peroxide(H2O2),and activities of superoxide dismutase(SOD),glutathione reductase(GR)and catalase(CAT)were studied by leaf segment method in vitro.[Result] After the treatment of 500 μmol/L TeA,the content of MDA and H2O2 increased by 247.86% and 67.00%,respectively,indicating that the accumulation of MDA and H2O2 in E.crus-galli leaves was due to the reactive oxygen burst induced by TeA.TeA induced a significant increase in activities of SOD,GR and CAT.At 500 μmol/L TeA,activities of SOD,GR and CAT increased more than one fold compared with the control.[Conclusion] TeA could not only cause oxidative stress in leaves of E.crus-galli through the induction of reactive oxygen,but also induce the increasing of antioxidant enzyme activity.
