不同基质与生长素处理对长白山笃斯越桔嫩枝扦插生根的影响被引量:10

以长白山笃斯越桔2年实生苗木嫩枝为试材,进行扦插基质、生长素种类和最佳处理浓度筛选试验。结果表明:长白山笃斯越桔以草炭土为基质,用200mg/L IBA浸泡处理的生根质量最佳,500mg/L生根粉速蘸的生根质量亦可。

长白山区3种乌头属植物对3种农作物螟虫触杀活性的比较被引量:8

[目的]基于均匀设计法探讨黄花乌头、北乌头和高山乌头的水蒸气蒸馏液对大螟、玉米螟和大豆荚螟的触杀作用。[结果]对大螟的24 h触杀结果表明:3种乌头对大螟均有较强触杀作用,触杀活性顺序为北乌头>黄花乌头>高山乌头。48 h时黄花乌头和北乌头对大螟保持着较好的触杀作用,触杀活性顺序为北乌头>黄花乌头。72 h仅北乌头还保持着较强的触杀作用。24、48、72 h致死中浓度LC50值分别为12.72、5.43、1.44 mg/L,校正死亡率分别达85.7%、88.9%、98.2%;对玉米螟的24、48 h触杀结果表明:北乌头和高山乌头对玉米螟均有较强的触杀作用,触杀活性顺序为北乌头>高山乌头。72 h仅北乌头保持着较强的触杀作用。24、48、72 h致死中浓度LC50值分别为15.97、15.17、1.71 mg/L,校正死亡率分别达92.3%、95.0%、99.4%;对大豆荚螟的24、48、72 h触杀结果表明:北乌头和高山乌头对大豆荚螟均有较强的触杀作用,触杀活性顺序均为北乌头>高山乌头。其24、48、72 h致死中浓度LC50值分别为17.27、17.18、12.39 mg/L,校正死亡率分别达90.0%、91.8%、99.0%。[结论]3种乌头对大螟均有较强触杀作用,北乌头和高山乌头对玉米螟和大豆荚螟表现出较强的触杀效果,黄花乌头对玉米螟和大豆荚螟的触杀效果不显著。

Interspecific Cross Compatibility of Rhododendron in Changbai Mountain被引量:3

In order to explore the interspecific cross compatibility of Rhododendron and the feasibility of interspecific cross in germplasrn innovation, reciprocal crosses among five species of Rhododendron in Changbai Mountain were made, In addition, the hybrid embryos were cultured in vitro. The results showed that the species of Rhododendron in Changbai Mountain had significant differences in reciprocal cross compatibility. When Rh. chrysanthum Pall. was used as female parent, it showed high cross compatibilities with Rh. mucronulatum Turcz. (92.55%), Rh. mucronulamm Turcz. f, album Nakai (94.14%) and Yabuli Rhododendron (92.42%), and no signifi- cant differences were found in fruit setting rate by serf-poUination. The germination rates of hybrid embryos were 13.60%, 11.59% and 4.12%, respectively. The hybrid seedlings had weak growth vigor and albino status and were trended to be aborted. After 2-3 generations of subculture, the growth vigor of hybrid seedlings were re- covered gradually, and their differentiation rate was also significantly higher than that of Rh. chrysanthum Pall. seedlings. But the hybrid seedlings were still yellowish green. In reciprocal crosses, the fruit setting rates were significantly reduced. The combination with the highest fruit setting rate (36.51%) was Yabuli Rhododendron x Rh chrysanthum Pall The fruit setting rates of reciprocal crosses between Rh chrysan- thum Pall and Rh. mucronulatum Turcz. and Rh.: chrysanthum Pall. and Rh. schlip- penbachii Maxim were 6.52% and 0, respectively. However, the hybrid embryo's germination rate (39,81%) of Yabuli Rhododendron x Rh. chrysanthum Pall. was significantly higher than that of their original cross. Moreover, the hybrid seedlings grew strongly and could be directly transplanted without subculture.

天目铁木愈伤组织和芽苗诱导技术被引量:5

以天目铁木嫩茎尖为外植体,应用均匀设计法筛选其基部愈伤组织诱导和愈伤组织再分化芽苗的最适合培养基.结果表明,最适合的嫩茎尖基部愈伤组织诱导培养基为1/2DR+TDZ 2.30 mg.L-1+2,4-D 0.55mg.L-1,诱导率为93.5%;愈伤组织芽苗再分化培养基为1/2DR+TDZ 3.30mg.L-1+KT 0.70mg.L-1,分化率达99.8%以上.成功建立了天目铁木嫩茎尖离体诱导愈伤组织和芽苗再分化体系,且再生芽苗与野生植株染色体数目相同.

草莓试管内诱导匍匐茎和高温处理结合茎尖培养脱毒技术研究被引量:24

【目的】获得可用于生产的无病毒草莓种苗。【方法】在试管内对深山草莓花瓣愈伤组织分化产生的优良变异新品种的试管苗进行匍匐茎诱导,并采取试管内高温处理结合茎尖培养对该品种进行脱毒研究,应用均匀设计法对各步骤主要影响因子进行优化筛选。【结果】该草莓新品种试管苗匍匐茎诱导的适宜培养基为:LS+6-BA1.00mg/L+GA31.90mg/L+KT1.00mg/L,其诱导率为99.5%以上;将含有匍匐茎的试管苗在试管内于56℃条件下培养85d后,切取1mm的茎尖在培养基MS+6-BA0.50mg/L+NAA0.20mg/L中进行茎尖愈伤组织诱导,然后将茎尖愈伤组织转接到培养基MS+6-BA1.00mg/L+NAA0.05mg/L中进行愈伤组织芽苗再分化培养,对再生芽苗进行病毒检测,脱毒率为100%。【结论】利用试管内高温处理结合茎尖脱毒的方法可以完全脱除草莓病毒,从而建立了草莓脱毒体系。

Study on Multiplication, Rooting and Transplanting of Tissue Culture Plantlets of Rhododendron chrysanthum Pall

[Objective] This study aimed to shorten the multiplication culture and root- ing culture periods of Rh. chrysanthum Pall. [Method] The Rh. chrysanthum Pall tis- sue culture plantlets collected from Changbai Mountain were used as material, and the effects of different hormone combinations and coconut milk on the proliferation and differentiation of Rh. chrysanthum Pall tissue culture plantlets were investigated. In addition, the rooting medium and transplanting matrix for Rh. chrysanthum Pall tissue culture plantlets were explored. [Result] The medium composed of modified MS, iBA (3 mg/L) and ZT (1.5 mg/L) was the optimum medium for subculture mul- tiplication of Rh. chrysanthum Pall tissue culture plantlets. The multiplication multiple and average plant height were significantly improved by adding coconut milk into the medium (150 mg/L). [Conclusion] For Rh. chrysanthum Pall tissue culture plantlets, the optimum rooting culture medium was composed of modified MS (1/4) and IBA (5.0 mg/L), and the tissue culture plantlets began to root 8 d after the inoculation. The root induction treatment was carried out after a 15-d sand culture, and the suitable matrix was composed of tufty soil, humus soil and perlite (2:1:1) with a survival rate of 95.66%.

羊踯躅嫩茎离体培养和植株再生被引量:3

本研究以羊踯躅嫩茎为外植体,应用均匀设计法筛选其最适合的嫩茎基部直接再生芽苗和生根培养基。结果表明,最适合羊踯躅嫩茎基部直接再生芽苗的诱导培养基为:1/4DR+2ip3.75 mg/L+IAA 0.03 mg/L+KT 1.30 mg/L,诱导率为99.8%;生根培养基为:1/4DR+ZT 0.06 mg/L+NAA 0.02 mg/L,生根率达99.0%以上。以再生植株茎节为材料进行快繁的结果表明,在28 d的培养周期内,每段增殖倍数平均达5以上。成功建立了羊踯躅的离体培养和植株再生体系,所建立的植株再生体系可以满足羊踯躅工厂化育苗的需要。

深山草莓花瓣离体培养诱导变异株系研究被引量:3

应用均匀设计法探讨了影响深山草莓花瓣愈伤组织诱导及愈伤组织再分化变异植株的主要因子和水平,并从中获得稳定的变异株系。结果显示:深山草莓花瓣愈伤组织诱导率超过65.0%以上的培养基共有11个,其中以LS+TDZ 2.06 mg.L-1+2,4-D 0.60 mg.L-1培养基的诱导效果最好,诱导率达98.0%;最佳再分化培养基为1/2LS+TDZ 2.30 mg.L-1+KT 1.38mg.L-1,田间试验发现变异率高达2.77%。再生植株生根移栽后观察筛选获得6个性状表现稳定的变异株系,再经过7年的栽培试验获得了1个性状稳定的优良变异株系。

以烈香杜鹃嫩茎为外植体,应用均匀设计法筛选其最适合的嫩茎基部直接再生芽苗和生根培养基。结果表明,最适合烈香杜鹃嫩茎基部直接再生芽苗的诱导培养基为DR+2·30mg·L-1TDZ+0·05mg·L-1IAA+1·80mg·L-1KT,诱导率为99·6%;生根培养基为1/2DR+0·07mg·L-1IAA+0·02mg·L-1NAA,生根率达99·7%以上。以再生植株茎节为材料进行快速繁殖,在35d的培养周期内,每段增殖倍数平均达5以上。

【目的】建立烈香杜鹃嫩茎段腋芽丛生芽诱导和植株再生体系,为烈香杜鹃工厂化育苗奠定技术基础。【方法】以烈香杜鹃嫩茎为外植体,应用均匀设计法对影响烈香杜鹃腋芽丛生芽诱导和生根的各主要植物生长调节剂及其水平的作用进行探讨。【结果】适宜的烈香杜鹃嫩茎段腋芽丛生芽诱导培养基为:1/2DR+TDZ 2.80mg/L+IAA 0.01mg/L+GA31.85mg/L,诱导率为99.8%;生根培养基为:1/2DR+IAA 0.07mg/L+NAA 0.02mg/L,生根率达99.7%。在生根培养基中添加GA32.00mg/L,再生植株茎节在35d的培养周期内,每段增殖倍数平均达5以上,植株再生率为97.0%。再生植株炼苗移培后成活率达95.0%以上。【结论】建立了烈香杜鹃嫩茎段腋芽丛生芽植株再生体系。
