Adenovirus F40/41 Gastroenteritis-Associated Emphysematous Gastritis:A Case Report

Emphysematous gastritis(EG)is a rare form of gastritis characterized by gastric wall pneumatosis.It is associated with a high mortality rate.Our case report describes a patient with a history of atrial fibrillation treated with apixaban,prior stroke,and schizoaffective disorder who showed epigastric tenderness on examination.However,no peritonitis signs were recorded,and laboratory values revealed high lactate levels and the presence of adenovirus F40/41.Abdominal computed tomography scan showed extensive portal venous gas in the left hepatic lobe of the liver,as well as pneumatosis involving the entire stomach,indicating EG.The diagnosis was histologically confirmed and initially managed medically,which proved inefficacious,necessitating surgical intervention in the form of a laparotomy and esophagojejunostomy with Roux-en-Y reconstruction.This case report suggests a potential association between EG and adenovirus F40/41–induced gastroenteritis,thereby emphasizing the challenging nature of EG.It underscores the need for comprehensive research to enhance our understanding of its underlying causes and effective treatment strategies.

GAPDH suppresses adenovirus-induced oxidative stress and enables a superfast production of recombinant adenovirus

Recombinant adenovirus(rAdV)is a commonly used vector system for gene transfer.Efficient initial packaging and subsequent production of rAdV remains time-consuming and labor-intensive,possibly attributable to rAdv infection-associated oxidative stress and reactive oxygen species(ROS)production.Here,we show that exogenous GAPDH expression mitigates adenovirus-induced ROS-associated apoptosis in HEK293 cells,and expedites adenovirus production.By stably overexpressing GAPDH in HEK293(293G)and 293pTP(293GP)cells,respectively,we demonstrated that rAdV-induced RoS production and cell apoptosis were significantly suppressed in 293G and 293GP cells.Transfection of 293G cells with adenoviral plasmid pAd-G2Luc yielded much higher titers of Ad-G2Luc at day 7 than that in HEK293 cells.Similarly,Ad-G2Luc was amplified more efficiently in 293G than in HEK293 cells.We further showed that transfection of 293GP cells with pAd-G2Luc produced much higher titers of Ad-G2Luc at day 5 than that of 293pTP cells.293GP cells amplified the Ad-G2Luc much more efficiently than 293pTP cells,indicating that exogenous GAPDH can further augment pTP-enhanced adenovirus production.These results demonstrate that exogenous GAPDH can effectively suppress adenovirus-induced ROS and thus accelerate adenovirus production.Therefore,the engineered 293GP cells represent a superfast rAdV production system for adenovirus-based gene transfer and gene therapy.

腺病毒肺炎患儿干扰素-λ1表达水平与腺病毒载量的关系

目的:研究人腺病毒(human adenovirus,HAdV)感染后呼吸道上皮细胞中的λ1干扰素(interferon-λ1,IFN-λ1)的表达情况。方法:选取2019年1―12月因腺病毒肺炎住院的143例患儿为HAdV组,另选取同期35例呼吸道病原结果均阴性者作对照组,采集鼻咽拭子提取核酸,用荧光定量PCR检测病毒载量、IFN-λ1 mRNA表达水平和HAdV测序分型。结果:HAdV组、HAdV组中不同年龄段、单纯感染组、混合感染组、重症组、非重症组、HAdV-3组、HAdV-7组IFN-λ1 mRNA的表达水平均低于对照组。重症组HAdV载量高于非重症组;HAdV组、重症组、HAdV-7组的IFN-λ1 mRNA的表达水平与病毒载量有相关性。结论:儿童腺病毒肺炎病情严重程度与病毒载量相关,其IFN-λ1表达水平降低,提示IFN-λ1可能参与了腺病毒感染的疾病进程。

Detection of Adenovirus in Fresh Fruit, Vegetables, Wastewater and Manure from Irrigated Farms in Ouagadougou, Burkina Faso

Enteric viral pathogens are responsible for numerous epidemics associated with the consumption of fresh fruit and vegetable, whether raw or minimally processed. The aim of the present study was to assess agricultural practices and the presence of adenovirus (AdV) in fruits and vegetables, manure and irrigation wastewater sampled in the urban and peri-urban perimeters of Ouagadougou. A total of 286 samples including 30 lettuces, 42 tomatoes, 30 carrots, 30 strawberries, 74 manures and 80 wastewater samples were collected from four market garden sites in and around Ouagadougou. Nested PCR was performed with specific primers to detect adenoviruses (AdVs). A face-to-face survey was carried out using a questionnaire on market garden production practices. Overall, adenoviruses prevalence was 5.9% [IC95, 3.2% - 8.7%] in all samples analyzed. It was specifically 7.14% (3/42) from tomatoes, 6.7% (2/30) from lettuces, 20% (6/30) on strawberries and 7.5% (6/80) in irrigation water. The survey showed that irrigation water came from untreated sources (dam, well, canal) and then 52% of farms used untreated manure. No farms have implemented measures to limit access by domestic and wild animals. This work shows the presence of human adenoviruses in surface irrigation water and fresh produce, which is of concern when fresh produce is consumed raw. To reduce the public health risks associated with consuming these foods, it is essential to follow good hygiene and cultivation practices.

一种溶瘤腺病毒抑制肿瘤细胞的作用机制及特异性研究

研究重组人5型腺病毒(H101)抑制肿瘤细胞增殖的作用机制及特异性。采用体外细胞培养方法及TCID_(50)检测病毒滴度法,发现H101对p53^(−)的肿瘤细胞有显著的选择性复制作用,其活性可以对p53突变的肿瘤组织产生溶瘤作用从而达到治疗效果;采用体外药效学实验检测H101对10种肿瘤细胞株的抑增殖作用,通过计算IC_(50)、IC_(90)进一步得出小鼠肝癌细胞Hepa1-6、小鼠胰腺癌细胞Pan02、小鼠神经胶质瘤细胞GL261对H101较为敏感。研究结果表明H101对已获批的适应证之外的其他肿瘤细胞也有溶瘤作用,具有适应证扩展的潜力。本研究可为后续开展动物模型的体内药效实验和使用剂量的选择提供参考。

重组腺病毒5型-诺如病毒GⅡ.4型-VP1病毒样颗粒的表达及鉴定

目的 采用293T细胞表达重组腺病毒5型(recombinant adenovirus 5,rAd5)-诺如病毒(Norovirus,NoV)GⅡ.4型-VP1病毒样颗粒(virus-like particle,VLP),并进行鉴定。方法 将重组腺病毒质粒pAd5-eGFP及pAd5-NoV-GⅡ.4-VP1分别转染293T细胞,拯救获得重组腺病毒rAd5-eGFP和rAd5-NoV-GⅡ.4-VP1。rAd5-eGFP在293T细胞上连续传代,验证其载体功能。将rAd5-NoV-GⅡ.4-VP1在293T细胞上进行传代,NoV-GⅡ.4-VP1经表达及纯化后,自组装形成NoV-GⅡ.4-VLP,并进行Western blot、ELISA检测及电镜观察。结果 镜下观察各代rAd5-eGFP均可见绿色荧光,且亮度随代次增加而增强。各代rAd5-NoV-GⅡ.4-VP1收获液中均可见NoV-GⅡ.4-VP1蛋白表达,相对分子质量约58 900。NoV-GⅡ.4-VLP可与兔抗NoV-GⅡ.4-VP1血清发生特异性结合,具有与天然的NoV病毒颗粒相似构象,可有效识别志愿者唾液样本中的NoV受体;镜下观察,其形态完整,呈球状,直径为43.5~58.3 nm,颗粒表面有少数突起,可能为VP1自组装时暴露于颗粒表面的P结构域。结论 表达的重组腺病毒NoV-GⅡ.4-VLP具有完整的VLP结构和良好的特异性,有望用于NoV腺病毒载体疫苗的相关研究。

儿童重症腺病毒肺炎

人腺病毒作为儿童社区获得性肺炎的重要病原,是重症肺炎的常见病原之一,其较高的致死率及长期后遗症发生率对患儿家庭及社会造成巨大负担。关注重症腺病毒肺炎的危险因素,包括病毒型别及载量、患者年龄及免疫状态、环境因素等。早期诊断和综合治疗有助于改善预后。本文就重症腺病毒肺炎的损伤机制、危险因素、临床表现及诊疗和预后等进行阐述。

西多福韦在腺病毒感染治疗中的应用

腺病毒是一种双链DNA病毒,可引起呼吸道、胃肠道、眼睛、肾脏和其他器官感染,轻度腺病毒感染一般是自限性疾病,但部分患者进展为高热、呼吸困难、呼吸衰竭等严重感染或播散性感染疾病。目前尚无特异性抗腺病毒药物上市,本文总结了西多福韦在腺病毒感染临床治疗中的应用,从成人重症腺病毒感染、儿童腺病毒感染、器官移植患者感染及衍生物4个方面展开阐述。总体而言,西多福韦在治疗腺病毒重症感染中有一定作用,但多数研究样本量偏少且缺少前瞻性队列研究,今后开展相关临床研究时需重点考虑。另外基于西多福韦开发衍生化合物或寻找类似药物,降低其肾毒性,也是未来研究关注的重点。

人腺病毒感染治疗的研究进展被引量:1

人腺病毒传染性强,可引起多个器官病变,部分人群感染后可发展为危重症,预后差,病死率高。目前临床上使用的抗腺病毒药物的疗效和安全性存在争议。近年来,越来越多的化合物和中药在细胞或动物实验中显现出显著的抗腺病毒活性及良好的安全性。此外,T细胞免疫疗法、抗病毒单克隆抗体疗法和腺病毒疫苗也被证实可有效对抗腺病毒感染。本综述对抗腺病毒领域的最新研究和进展进行总结,为新型抗腺病毒药物的研制和临床工作提供参考借鉴。

一株血清8a型禽腺病毒的分离鉴定及致病性分析

【目的】研究福建南平地区禽腺病毒(FAdV)血清8a型分离株的遗传演化情况及致病性,旨在为防控血清8a型FAdV感染提供参考。【方法】2022年8月,采集福建南平地区部分肉鸡养殖场出现的以包涵体肝炎为特征的肝脏病料,采用PCR技术检测FAdV核酸,将获得的阳性样品接种在无特定病原体(SPF)鸡胚上进行病原的分离纯化,利用hexon基因测序及序列分析等方法进行病毒鉴定,并在测定分离株毒力的基础上进行动物致病性试验。【结果】接种10 d后,鸡胚死亡,肝脏肿大、出血、坏死、呈土黄色;PCR鉴定和hexon基因序列分析表明,分离株与GenBank上的FAdV E型8a血清型澳大利亚TR59毒株的序列同源性高达99.5%,将该分离株命名为FJNP。FJNP株对1日龄SPF鸡的致死率为44%(11/25);剖检显示,感染鸡肝脏肿大、出血、边缘钝圆、呈土黄色、出现白色坏死点,脾脏和肾脏也出现肿大、出血等症状;肝脏组织病理学检查结果显示,感染鸡肝细胞大量变性坏死,肝细胞核可见嗜碱性包涵体及大量淋巴细胞浸润。实时荧光定量PCR检测显示,感染鸡体内多个组织器官均检测到病毒,且主要分布在肝脏,攻毒3、5、7、14、21 d在泄殖腔中均能检测到病毒。【结论】本研究从临床表现为包涵体肝炎的病鸡肝脏病料中分离到一株具有较强致病性的血清8a型FAdV。
